Design Notes
The part is designed to contained regions homologous to the psaD gene. As such we had to ensure that the homologous region was of sufficient length to allow for recombination. The part contains two regions homologous to psaD: the first region is homologous to the first 250 bases of psaD, the second region to the last 250 bases of the psaD gene. The centre of the part contains a kanamycin resistance gene, which becomes inserted in the middle of the chromosomal psaD gene during recombination.
